Apoptosis: A Target for Anticancer Therapy

Abstract
Apoptosis, the cell’s natural mechanism for death, is a promising target for anticancer therapy. Both the intrinsic and extrinsic pathways use caspases to carry out apoptosis through the cleavage of hundreds of proteins. In cancer, the apoptotic pathway is typically inhibited through a wide variety of means including overexpression of antiapoptotic proteins and under-expression of proapoptotic proteins. Many of these changes cause intrinsic resistance to the most common anticancer therapy, chemotherapy. Promising new anticancer therapies are plant-derived compounds that exhibit anticancer activity through activating the apoptotic pathway.
10. Plant-Derived Compounds Exhibiting Anti-Cancerous Activity
In light of increasing resistance to chemo- and radiotherapy along with the toxicity of these traditional therapies, a new, non-toxic anticancer treatment is needed. Additionally, chemotherapeutic agents are toxic to both normal cells and tumor cells. The optimal therapy would be to be able to differentiate between the cell types. Plant-based or -derived compounds are typically non-toxic to normal cells. For over 5000 years, plants have been utilized as medicines and therapies; a quarter of all modern medicine is directly or indirectly derived from plants [25].
Graviola is a fruit tree that has been used in both alternative and traditional medicine for a wide variety of aliments [26], particularly for its anticancer properties. It has been shown to inhibit BCL-2 proteins while increasing BAX and promoting apoptosis [27]. The mechanism graviola uses is still unknown, however, it is a potential anticancer treatment due to its nontoxicity towards healthy cells [26]. This makes graviola an exciting new possible therapy especially in comparison to the current treatments of chemo- and radiotherapy...
There are many other plant-derived compounds that induce apoptosis in cancer cells (Table 1). These include black cohosh of Actaea racemosa [28], Juglone from Juglans mandshurica [29] and genistein [30]. Quercetin, which is found in apples and red onions, activates caspases which leads to the apoptotic response [30]. Green tea is also thought to induce apoptosis in cancer cells, specifically through the compound epigallocatechin-3-gallate [31]. Aloe-emodin, found in Rheum palmatum, also has caspase-activation activity in cancer cells [31]. All of these plant-derived compounds have yet to be fully investigated but are noteworthy compounds that could be the future of cancer therapy. The nontoxicity towards normal, healthy cells while still attacking and producing an apoptotic response in cancer cells make these compounds particularly intriguing.

Abstract
Magnetic fields are commonly used in therapies designed for subjects with rheumatic diseases, yet the effects of magnetotherapy are not entirely clear in these disorders.
This study is designed to examine the literature investigating applications of magnetotherapy in the treatment of rheumatoid arthritis (RA).
The review focused on publications related to administering magnetotherapy in patients with RA. The databases Science Direct, SpringerLink, Medline, PubMed, and Polska Bibliografia Lekarska were searched for reports published since 2005.
Despite the numerous reports showing an impact of magnetic field in subjects with RA, the effectiveness of magnetotherapy has not been explicitly confirmed. Given the above, further research appears to be necessary to clarify the impact of magnetic fields on biological systems, and the relationship between magnetic field intensity and the obtained results as well as their durability. The majority of clinical trials have failed to identify any undesirable outcomes or side effects of this physical therapeutic factor.

Two Common Over-the-Counter Compounds Reduce COVID-19 Virus Replication by 99% in Early Testing
A pair of over-the-counter compounds has been found in preliminary tests to inhibit the virus that causes COVID-19, University of Florida Health researchers have found.
The combination includes diphenhydramine, an antihistamine used for allergy symptoms. When paired with lactoferrin, a protein found in cow and human milk, the compounds were found to hinder the SARS-CoV-2 virus during tests in monkey cells and human lung cells.
The findings by David A. Ostrov, Ph.D., an immunologist and associate professor in the UF College of Medicine’s department of pathology, immunology and laboratory medicine and his colleagues, are published in the journal Pathogens.
“We found out why certain drugs are active against the virus that causes COVID-19. Then, we found an antiviral combination that can be effective, economical, and has a long history of safety,” Ostrov said.

Due to his earlier research with colleagues at UF, Ostrov already knew diphenhydramine was potentially effective against the SARS-CoV-2 virus. The latest discovery has its roots in a routine meeting of scientists with the Global Virus Network’s COVID-19 task force. One researcher presented unpublished data on federally approved compounds that inhibit SARS-CoV-2 activity, including lactoferrin.
Like diphenhydramine, lactoferrin is available without a prescription. Ostrov thought about pairing it with diphenhydramine and ran with the idea. In lab tests on human and monkey cells, the combination was particularly potent: Individually, the two compounds each inhibited SARS-CoV-2 virus replication by about 30%. Together, they reduced virus replication by 99%.
The findings, Ostrov said, are a first step in developing a formulation that could be used to accelerate COVID-19 recovery. It also raises the prospect of further study through an academic-corporate partnership for human clinical trials focused on COVID-19 prevention. Additional research into the compounds’ effectiveness for COVID-19 prevention is already underway in mouse models.
To establish their findings, the research team focused on proteins expressed in human cells known as sigma receptors. In COVID-19 cases, the virus “hijacks” stress-response machinery, including sigma receptors, in order to replicate in the body. Interfering with that signaling appears to be the key to inhibiting the virus’s potency. “We now know the detailed mechanism of how certain drugs inhibit SARS-CoV-2 infection,” Ostrov said.
Data from the experiments show that a highly specific sigma receptor binding drug candidate (with pain relieving properties), and formulated combinations of over-the-counter products (such as diphenhydramine and lactoferrin) have the potential to inhibit virus infection and decrease recovery time from COVID-19, the researchers concluded.
While the findings are encouraging, Ostrov cautions against self-medicating with either diphenhydramine or lactoferrin as a COVID-19 prevention or treatment. The type of lactoferrin used in the research differs slightly from the type that is commonly available to consumers, he noted. Lactoferrin is commonly used as a supplement to treat stomach and intestinal ulcers, among other uses.

(1) Background: There is a strong need for prevention and treatment strategies for COVID-19 that are not impacted by SARS-CoV-2 mutations emerging in variants of concern. After virus infection, host ER resident sigma receptors form direct interactions with non-structural SARS-CoV-2 proteins present in the replication complex. (2) Methods: In this work, highly specific sigma receptor ligands were investigated for their ability to inhibit both SARS-CoV-2 genome replication and virus induced cellular toxicity. This study found antiviral activity associated with agonism of the sigma-1 receptor (e.g., SA4503), ligation of the sigma-2 receptor (e.g., CM398), and a combination of the two pathways (e.g., AZ66). (3) Results: Intermolecular contacts between these ligands and sigma receptors were identified by structural modeling. (4) Conclusions: Sigma receptor ligands and drugs with off-target sigma receptor binding characteristics were effective at inhibiting SARS-CoV-2 infection in primate and human cells, representing a potential therapeutic avenue for COVID-19 prevention and treatment

As I first reported on February 9th 2020, and again on March 15th 2020, researchers at the Indian Institute of Technology in Delhi had published a paper on January 31st about their findings of four unique insertions in Novel Coronavirus. These were the features which made it “novel”, because they had not previously been found in any other coronavirus.
It just so happens that these insertions appeared to be the same spike glycoprotein GP120 found on the HIV virus. In the Abstract, the Indian scientists wrote that this fact was “unlikely to be fortuitous in nature,” which was a delicate way of saying that SARS CoV-2 is a bioweapon.
The four HIV GP120 and gag protein receptor sequences that they’d discovered in the SARS CoV-2 virus are what allow both the HIV and COVID viruses to enter the cells of human bodies and to bind to human CD4 (T type white blood) cells.
If the presence of HIV GP120 in the Novel Coronavirus were due to random mutation/recombination, we’d expect to see other HIV sequences randomly distributed in the Wuhan virus’s genome. But we don’t.
These HIV sequences are found ONLY in critical binding sites, which is a clear indication of intentionality.
The Indian scientists were swiftly forced to withdraw their paper and The Lancet immediately published a statement signed by 27 scientists completely rejecting the possibility that the virus may have leaked from the Wuhan Institute of Virology, which effectively killed any meaningful study into the origin of the virus.
Nearly two years later, we’ve come full circle.
Biotech analyst and former Pfizer employee, Karen Kingston joins Stew Peters to discuss how many of those injected with the so-called “vaccine” are having their immune systems wiped out and they’re exhibiting rare symptoms that are commonly seen in AIDS patients, such as Kaposi’s sarcoma-associated herpesvirus, Mycobacterium avium complex [MAC] and depleted CD4 counts.
She then gets into the evidence of how the vaccine produces the spike protein that contains HIV GP120, which could explain why these unusual symptoms are happening.
She says, “The codon, the sequence for HIV glycoprotein 120 is in the spike protein. It’s called ‘double CGG’…In June of this year, the Wall Street Journal did an article on this and what they said was, ‘the double CGG is suppressed naturally, the opposite is true in laboratory work. The insertion sequence of choice is the double CGG.’
“What does this mean? It means it’s not a naturally-occurring genetic code and scientists put it into chimeric gain-of-function viruses intentionally for two reasons. 1) They know then, they marked it with their code and 2) they can track it.
“But what’s more alarming is that when the spike glycoprotein was released, it was actually sequenced by a number of scientists in India in January of this year [sic – it was in 2020] and they found four insertions of HIV in the spike glycoprotein. Four. And HIV spike glycoprotein 120, what it does is it completely depletes your CD4 cells.
“And what’s more alarming is…one study that was done in 2004 and you see the usual suspects: an NIH-sponsored study, it’s done with folks at Harvard, it’s done with an AIDS research organization, it’s done with Berlin and it’s done out of Peking University in China and what they did was they took coronavirus SARS-CoV-1 and they wanted to optimize it for infection and receptor to the ACE-2 inhibitor.
“And the way that they did it was they put monkey HIV, simian HIV in there, they used murine mouse leukemia codons. But what’s most interesting is they said: ‘Infection mediation by a spike protein variant whose cytoplasmic domain had been truncated and altered to include a fragment of the cytoplasmic tail of the Human Immunodeficiency Virus Type 1 envelope glycoprotein was, in both cases more efficient than the wild-type spike protein.’
“So by adding HIV glycoprotein 120, they made it more receptive to the ACE-2 receptors in your lungs and kidneys, as well as to your T cells, your CD4 cells, to annihilate them.
“Now, this is the part where people’s jaws are going to drop: We know CGG is not found in nature, therefore, it’s patentable. Well, who did all the research on this? And who owns it? Well, Dr Fauci did a number of studies, he paid himself over $11.677 million with US Taxpayer dollars, between 2014 and 2020 to research, using glycoprotein 120 from HIV and integrating it into coronaviruses – and he patented it. He owns several patents on this.
“So every time someone’s being injected with the synthetic mRNA, producing the Wuhan-HU-1 spike glycoprotein, Fauci is making a dime…

We are currently witnessing a major epidemic caused by the 2019 novel coronavirus (2019-nCoV). The evolution of 2019-nCoV remains elusive. We found 4 insertions in the spike glycoprotein (S) which are unique to the 2019-nCoV and are not present in other coronaviruses. Importantly, amino acid residues in all the 4 inserts have identity or similarity to those in the HIV-1 gp120 or HIV-1 Gag. Interestingly, despite the inserts being discontinuous on the primary amino acid sequence, 3D-modelling of the 2019-nCoV suggests that they converge to constitute the receptor binding site. The finding of 4 unique inserts in the 2019-nCoV, all of which have identity /similarity to amino acid residues in key structural proteins of HIV-1 is unlikely to be fortuitous in nature. This work provides yet unknown insights on 2019-nCoV and sheds light on the evolution and pathogenicity of this virus with important implications for diagnosis of this virus.

Severe acute respiratory syndrome coronavirus 2 (SARS–CoV–2) has led to the coronavirus disease 2019 (COVID–19) pandemic, severely affecting public health and the global economy. Adaptive immunity plays a crucial role in fighting against SARS–CoV–2 infection and directly influences the clinical outcomes of patients. Clinical studies have indicated that patients with severe COVID–19 exhibit delayed and weak adaptive immune responses; however, the mechanism by which SARS–CoV–2 impedes adaptive immunity remains unclear. Here, by using an in vitro cell line, we report that the SARS–CoV–2 spike protein significantly inhibits DNA damage repair, which is required for effective V(D)J recombination in adaptive immunity. Mechanistically, we found that the spike protein localizes in the nucleus and inhibits DNA damage repair by impeding key DNA repair protein BRCA1 and 53BP1 recruitment to the damage site. Our findings reveal a potential molecular mechanism by which the spike protein might impede adaptive immunity and underscore the potential side effects of full-length spike-based vaccines.
